Life Changes – Halting Plasmid Distribution

As some of you may know I recently obtained my PhD 🙂 and have been looking for work. I’m very happy to say that a wonderful opportunity has come up! I will be starting work at a biotech company on the microfluidics / molecular biology side!

As I transition to this new non-academic world, I’m voluntarily deciding to halt distribution of the plasmid pack. I will fulfill requests made up to today (Feb 16th, 2021), however going forward I will have to decline further requests. This is to avoid any potential conflicts of interest and to focus myself for the tasks at hand.

The blog will continue to grow and mature, this is a labor of love and I appreciate meeting all the wonderful DIYers and scientists around the globe.

  1. I got the plasmid pack from Alex last year, I’d be happy to share Alex’s plasmids ( when I am still employed in University ) as long as I doesn’t cost me an arm or a leg for shipping.

Recently I had a researcher from UCLA organised FedEx label for me to ship the plasmid to them in USA.

    1. Hi Ryan

I would like to get the plasmid pack and try out the enzymes. I’ve been using my own clone of pfu for several years now, but recently I’m cloning a lot of long templates and my pfu is not working to the desired level. It would be great if you could send me the clones you obtained from Alex. I can give you my FedEx account number if it helps.

      Thanks!

      1. Dear Gabriel,

I just find this website today. And I’m wondering have you got the plasmid packs last year from Ryan? If yes, is it possible for you to send the plasmid packs to me? Thanks a lot in advance!

        Zimo

    2. Hello Ryan!
Have you purified Mashup RT? How enzyme performes?
On the site I found only amplification of Gapdh, which is not good control, cause even if one has 90% enzyme degradation, still will get amplification FROM GAPDH cdna, but not from any other..

      1. To everyone who sends me plasmid request, here is mail template:
        ” Dear Mariia,
Is it possible for you to share the plasmids (NAME)?
Here is my address for regular post:
        NAME/UNIVERSITY/street/country/POST code.
If you prefer FEDEX/DHL please send me your information and I will create shipping label.
        Thank you for your time”
Emails ” Hey, Send me please the plasmids because my lab is very poor, Regards.” I WIL IGNORE.
It took me 3 days and 4 mails to get correct post information for a last request. People everyone lets respect each others time

    3. Hi Ryan,

This might be a long shot, but would you be able to share some of Alex’s plasmids? I’d really appreciate it.

      Cheers,
      Ben

  2. I am able to ship out plasmids until November 2021, and expected to conclude my work at deakin university. Get in touch before that time (see my email in comments above) if you wish to get a plasmid pack.

In the spirit of paying it forward, it would be great if there’s any volunteer who could assist with distribution going forward.

    1. Hi Ryan, has anyone took over this task? I am wondering if there is anyone that share a little sample of MMLV RT plasmid with me.

    2. I have sent an e-mail to you for assisting distribution forward and a request for new plasmids but have not received a reply, so whatever the case I am willing to forward plasmids that people require for the future but I need to receive them first 🙂

      Take care,

      ML

  3. Hi all,

Would anyone be willing to share the plasmid pack? I would be super grateful if so and happy to pay for shipping!

  4. Hi Guys, I am curious that if I can get Alex’s plasmids pack, then I can ask people to request them from me and pass this sharing tradition from you to me and to other DIYers.

  5. Would anyone be willing to share the 3 plasmids: Sumo-MRI, T4 DNA ligase, DpnI to me, then other DIYer can request them from me and pass this sharing tradition forward.

  6. Hey everyone,

I’d also like the plasmid pack and can continue to expand and distribute once it’s received. Also, have a number of fluorescent proteins (BFP, CFP, GFP, YFP, CyOFP, dsRed) in the bacteria blue light-inducible pDawn vector “addgene.org/43796” to add to the collection. Please reply or reach out to me directly john_lamb(at)uml.edu to arrange shipping.

    Thanks,
    John

  7. In the spirit of being given some of the plasmids, I am also happy to share! You can e-mail me through your institutional e-mail (so I know that I will be sharing academically) to cihan.aydin at gmail with a FedEx (or equivalent) and I will send you a filter paper absorbed plasmid. However my schedule is quite erratic and I tend to forget stuff so please feel free to remind me!

So what I have to this date is…
    pML-MRI, pML-MashUp RT, pML-DpnI, p50-T4 Ligase, Pfu-sso7d

    What I did for myself…
In pColdI and pCold-TF -> HisTaq, Phusion (Codon Optimized), MashUp, p50-T4Lig

    What I seek…
    CTEV, MBP-DpnI, MBP-MRI

    Take care,

    ML

  8. Found your homepage today, great read, super helpful and would be great to get the plasmid pack from someone out there, can provide a fedex label if needed.

    Thanks in advance,
